When we receive a phone call on our Asterisk/Elastix server i want to route it automatically to the user to which the contact is assigned to in Vtiger. So when we receive a call in asterisk this should check of the number is connected to a contact in vtiger. When it is asterisk should check if the extension for this user is online and transfer call to him.

You should use vTiger API which will be integrated with Asterisk PHP-AGI installed on your Asterisk environment. This PHP-AGI will check the contact over its API and do the transfer once it found matched.
